Nope, it was first used in the movie and then for the show in season 5.

Nope infact Karen was barely even a character in pre movie

Even with Karen technically being around since Plankton's first appearance, she really didn't get a bigger role until season 3 when we actually learned her name. It was a good idea to switch to a mobile form so she can be less limited on what she can do.

I just want to say that Karen(design + character) is one of the only things I think post movie is better than pre movie

Technically yes and no. Her name was listed in the credits of "Jellyfishing/Plankton!" so her name was already established at that point but it was never actually said in-series until The Algae's Always Greener.
